Fate of Peripheral Nerve Transplantation in Rats

Abstract
Rat sciatic nerve segments (1.5 cm long) were subjected to iso-(Lewis-Lewis) and allografting (BN-Lewis) using 9-0 nylon microsutures by end-to-end epineural anastomosis. Push-off returned within 2 months in all animals allografted and 6 weeks in all isografted animals that followed. Animals were sacrificed at 6 to 12 hours, days 1, 3, 5, 7,12,14, 39, 60, 90 and 180, and the grafts were examined grossly and histologically. There were equal amounts of polymorphonuclear neutrophil cells at 6 hours and the grafts were indistinguishable during the 1st and 2nd days after grafting. After day 3, a mononuclear lymphocyte and histiocytic cellular infiltrate occurred and persisted for 14days in the allografts and then subsided by day 30. The mild inflammatory infiltrate always remained at the suture sites for 180 days. Both isoand allografts were indistinguishable by day 60 and thereafter. The size of the nerve graft may be important for survival even if nerve is subjected to lesser immunological attack than other tissues.
